RAPD method in examinations of hybrids of triticale and Aegilops juvenalis (Thell.) Eig

abstract: The aim of the study was estimation of genetic similarity in triticale hybrids using the RAPD method and identification of the wild species DNA in the triticale background. PCR was performed using 16 RAPD primers. A total number of 141 bands was obtained. Examined primers amplified between 5 (U572) and 14 (A16) DNA fragments. The average number of polymorphic product was 2.9 per primer – from 1 (for F05 and W2) to 6 (for U225). Three primers amplified 4 products identifying Aegilops juvenalis (Thell.) Eig chromatin in triticale background. The genetic similarity between the strains was estimated. The largest mean genetic similarity (0.846) to all examined genotypes was shown by Aegilops juvenalis 6x × [(Lanca × L506/79) × CZR142/79]. Mean similarity of all analysed forms was calculated at 0.791. On the basis of genetic similarity index matrix examined forms were divided into clusters by means of UPGMA method.
